 Operating Procedures

Before gel Casting, please ensure that the gel solution is ready and the glass plates are cleaned.

 

 

1.Firstly, place the gel casting clamp on a flat desk, fully open the pressure plate on the clamp (pressure plate state 1), insert the flat glass plate and concave glass plate (as shown in the following figure), ensure that the bottoms of the flat glass plate and concave glass plate are aligned, and rotate the pressure plate to tighten the glass plates. (pressure plate state 2)

Note: Before starting gel casting, check if the flat glass plate and concave glass plate are aligned vertically and horizontally, if the bottom of the glass plate is placed correctly, and if the pressure plates at both ends of the glass plate are tightened.

 

2. Gel Casting

a. Press the lower edge of the glass in the gel casting clamp onto the silicone base, clamp the upper edge of the concave glass plate with a clip to fix the glass plates. Slowly pour the prepared resolving gel (lower gel) solution into the gel chamber between the two glass plates until it reaches the scale line on the glass plate; (Servicebio glass plates come with scale lines, so pouring gel to the scale line ensures accuracy! For glass plates without scale lines, pour the solution to about 1.5-2.0cm from the upper edge of the flat glass plate)

b. If using the Servicebio one-step gel preparation kit (G2177), after pouring the lower gel, there is no need for liquid sealing, pressing flat the gel surface, or waiting for the resolving gel to solidify. You can directly pour the stacking gel (upper gel) solution and wait for gelation.


c. (Optional) Liquid sealing step: Use distilled water or absolute ethanol to evenly add left and right into the gel chamber to liquid seal and press flat the resolving gel (lower gel). Let it stand for about 20 minutes; after the gel solidifies, pour out the distilled water or absolute ethanol and use an absorbent paper to absorb any residual liquid in the gel chamber. Slowly pour the prepared stacking gel (upper gel) solution until the gel chamber is full, then immediately insert the electrophoresis comb. Let it stand for 15~20 minutes to allow the gel to solidify.

 

 

3.The gel casting stand can be used alone or combined into a whole as needed, effectively utilizing laboratory bench space.
